Coagulation cascade in sepsis: getting from bench to bedside?

چکیده

The relationship between blood coagulation factors and the promotion or inhibition of the antiinflammatory response continues to be defined through basic research. The potential key role of blood coagulation factors in the response during sepsis provides an exciting potential mechanism(s) for modification through the application of new therapies. The complexity of the potential multiple actions of the proteins, such as protein C, should allow for development of new therapies to minimize the detrimental inflammatory response. However, several gaps in our current understanding must be bridged before the clinician can take the basic knowledge ‘to the bedside’, where individual patients will benefit.

Bench to bedside: targeting coagulation and fibrinolysis in acute lung injury.

Substantial progress has been made in understanding the contribution of alterations in coagulation and fibrinolysis to the pathogenesis of acute lung injury (ALI). Findings from mouse, rat, baboon, and human studies indicate that alterations in coagulation and fibrinolysis may be of major pathogenetic importance in ALI and other inflammatory conditions in the lung including pneumonia, sepsis, a...

Bench-to-bedside review: Association of genetic variation with sepsis

Susceptibility and response to infectious disease is, in part, heritable. Initial attempts to identify the causal genetic polymorphisms have not been entirely successful because of the complexity of the genetic, epigenetic, and environmental factors that influence susceptibility and response to infectious disease and because of flaws in study design. Potential associations between clinical outc...

Promising role for Gc-MAF in cancer immunotherapy: from bench to bedside

Immunotherapy has been used for years in many types of cancer therapy. Recently, cancer immunotherapy has focused on mechanisms which can enhance the development of cell-mediated immunity. Anticancer medications are administered to inhibit immunosuppressive factors such as nagalase enzyme, which is produced by neoplastic cells and destroys macrophage activating factor (Gc-MAF). Anti-neoplastics...

Bench-to-bedside review: Circulating microparticles - a new player in sepsis?

In sepsis, inflammation and thrombosis are both the cause and the result of interactions between circulating (for example, leukocytes and platelets), endothelial and smooth muscle cells. Microparticles are proinflammatory and procoagulant fragments originating from plasma membrane generated after cellular activation and released in body fluids. In the vessel, they constitute a pool of bioactive...
